Disk Cleanup

Can anyone help, Disk cleanup will not run, I have run 3 different updated virus scan programs, have ran Spybot, Adaware, when I start disk cleanup it just sits there, I have let it sit there for an hour and it never runs, when I start it, it ties up my CPU 100 percent, and then I have to reboot to get it back to normal. Thanks for any help!
